Abstract

The present study was conducted to investigate the effect of diluent on freezing of Saanen and Damascus buck semen. Six mature bucks (3 Saanen and 3 damascus ) were used in this study to prepare semen samples which were collected with an artificial vagina. Sexual behaviour of the bucks was assessed by recording the reaction time. The study examined the sperm characteristics (volume of ejaculate, concentration, physical appearance, mass activity , sperm motility and morphology) Semen samples were diluted with skimmed milk, Triladyl and Andro Med. The diluted semen was preserved in liquid nitrogen at – 196c and examined for sperm motility after 48 hours. The results indicated that sexual desire was significantly affected by breed. Damascus bucks showed the strongest sexual desire (15.13 seconds), than Saanen bucks (26.41 seconds ). The results of semen characteristics were not significantly affected by breed except the concentration and abnormal sperms .Saanen bucks produced the greatest value (1.39 ml ) , while the smallest volume was recorded for Damascus, bucks (0.93ml ) sperm concentration has no significant affected Saanen bucks recorded (4.37 ×106 ) ,while Damascus bucks recorded (5.16×106 ). Mass activity of sperms showed significant affected, Damascus bucks recorded the higher score (2.33) than Saanen bucks (1.77). The percentage of sperms motility showed no significant differences between breeds. It was 68.72% in Damascus bucks and 65.45% in Saanen bucks. The lowest percentage of abnormal sperms was recorded in Saanen bucks (2.45%) and the highest in Damascus bucks (3.54%), but with no significane differences between breeds. The highest post-thawing motility was recorded in Damascus bucks for semen diluted with AndroMed (51.82%), and the lowest value was shown by semen diluted with Skimmed milk (42.08%). While in Saanen bucks the highest post-thawing motility of sperms was recorded by semen diluted with Triladyl (48.00%), the lowest value was encountered in semen diluted with AndroMed (40.00%).
