This study was carried out in Ed Babker area in Shergalnile-Khartoum State, during the period of March 2016 to September 2016. The objectives were to estimate the epidemiology and prevalence rate of plasmodium vivax, compare the prevalence rate of plasmodium falciparum, and compare the efficiency of blood films and polymerase chain reaction (PCR) as methods for diagnosing plasmodium species in blood inEd Babker area. A total number of 100 blood samples were collected from Ed Babker health center. All samples were examined by direct microscopy for P.malaria by blood film (thick and thin) and species of Plasmodium was identified for all positive samples. Then all samples were examined by nested PCR for confirmation. From 100 sample there were 73 (73%) samples negative, 17(17%) samples were positive for P.falciparum, 7(7%) samples were positive for P.vivax and 3(3%) samples were mixed (P.falciparum and P.vivax). From all positive samples, 12 samples positive by direct microscopy and PCR,and 2 samples were mixed. The study concluded that, the P.falciparum show the more common species in Shergalnile area. The PCR more sensitive than microscopy (p.value=000.0).en_US
