dc.description.abstractHuman papillomaviruses (HPV) have great public role in the development of cervical carcinoma causing fatal consequences. The aim of this study was to detect the presence of HPV-18 virus by amplification of the E6 gene by real time-polymerase chain reaction (rt-PCR) analysis utilizing the screening SYBER green. A total of 50 cervical cancer biopsy samples (parrafin embedded) of patients attending Khartoum Teaching Hospital, National Tropical and Health Laboratory, Army Medical Hospital and Soba ‫.‪countries‬‬ University Hospital were collected to be processed for further study. This study was conducted during the period from April to September 2012. The specimens were treated to remove paraffin, and DNA extracted. The E6 gene of HPV-18 encoding the oncoprotein product was amplified by the rt-PCR assay using the SYBER green. Out of the total 50, 18 (36%) specimens yielded positive results; resembling the sigmoid curve of the positive control, while 32 (64%) were negative, in addition most of the positive results were concentrated among age group (29-50 years) with frequency rate 12 (24%). In conclusion, this study showed that HPV-18 has great role in initiating cervical cancer with no noticeable variation on the occurrence of the virus in comparison with local and global studies in Sudan and other countries. Specimens were treated, paraffin removed and DNA was extracted and then E6 gene of HPV-18 was detected by real-time PCR.
