Human papillomavirus (HPV) is associated with cervical carcinoma causing high morbidity and mortality. The aim of this study was to detect HPV type-16 in cervical cancer tissue. A total of 50 cervical carcinoma biopsies (paraffin embedded and previously diagnosed as cervical carcinoma by histological examination) were collected from Khartoum Teaching Hospital, 3 National Tropical and Health Laboratory, Army Medical Hospital and Soba University Hospital. This study was performed during the period from April to September 2012. The specimens were treated and the paraffin removed, then the DNA was extracted followed by the detection of E6 gene of Human papillomavirus type-16 using non-probed SYBER green real time PCR. Out of the total 50, 30 (60%) showed positive results as compared with the sigmoid curve of the positive control for HPV-16; while 20 (40%) were negative. Most of the positive results concentrated among age group 31-50 years with 14 (28%) frequency rate. The study concluded that SYBER green rt-PCR technique detected more than 50% of the potential cervical cancerous tissues tested by the histological method. Further research applying probed rt-PCR method is warranted.en_US
