Abstract:
This a retrospective study was carried out the research laboratory, department of
Histopathology and Cytology in Sudan University of science and technology,
Khartoum Sudan. The objective of this study was to evaluate the effects of
Microwave oven in Melanin pigment staining procedures and to compare between
the classical Masson-Fontana staining technique at room temperature and
Microwave technique with Masson-Fontana stain. 50 skin tissue samples were
taken for processing. After processing, two histochemical staining techniques were
carried out ;classical Masson-Fontana method at room temperature and Microwave
oven Masson-Fontana staining method at temperature 20c. Both methods were
compared. The results of this study, indicated that the use of microwave oven in
staining of melanin pigment reduced the time from overnight to only 2minutes
with very good staining quality as compared of classical Masson-Fontana method.
Using Harris haemaoxylin and Eosin stain for histological changes examination
and diagnosis conformation. In conclusion, the use of microwave oven is a vital
tool for staining of melanin with Masson-Fontana method. Further research and
studies to improve the application of microwave oven in the staining of melanin
pigment with Masson-Fontana stain are recommended.
