Trials were conducted in the tissue culture laboratory of the Department of Tissue Culture, Commission of Biotechnology and Genetic Engineering, National Centre for Research, Khartoum, Sudan. The present study was undertaken to identify the best explant, salt formulation and salt-strength, sucrose concentration and the growth regulators that are adequate for direct shoot regeneration in fenugreek using shoot tips derived from in vitro germinated seedlings. Murashige and Skoog original medium was used as basal medium. Shoot tips gave the highest values in all growth parameters measured showing the greatest potential for response to in vitro growth and development compared to nodal stem segment explant. The results showed that both MS salts of the full or half strength were optimal for all parameters measured compared with other salt formulations and strengths tested, and there is no significant difference between them. Sucrose concentration of 2% was better than the other concentrations tested for growth and development of cultured shoot tips. The results revealed the superiority of BA to other cytokinin types tested. Best results were recorded on the nutrient medium containing 0.5 mg/l BA, where the greatest number of leaves, nodes and shoots were obtained. As for the combination treatments of NAA and BA the best results were registered with the combination of NAA at 0.0 mg/l and BA at 0.0mg/l where the largest number of leaves, nodes and shoots were obtained. Root formation occurred principally on basal media exclusive of growth2 regulators. |
