dc.description.abstract |
This study was conducted in Khartoum state the capital of Sudan, aimed to detect P-Selectin Glycoprotein Ligand-1 polymorphism among Sudanese pregnant women with deep vein thrombosis (DVT) in Third Trimester.
This is a case – control study included a total of 304 Sudanese pregnant women, 194(64%) Sudanese pregnant women in third trimester confirmed with DVT as a case group and 110 (36%) healthy pregnant women were recruited in the study as control group mean age 33 ± 3.67 for patients and 32 ± 3.7 mean age for control. Clinical, demographic and personal data were collected from predesigned questionnaire, Fibrinogen and D-dimer was performed from citrated blood samples, and Genomic DNA was isolated from whole EDTA blood, Genotyping of SELPG gene was performed by PCR amplification, electrophoresis and DNA sequencing. D-dimer results showed significant increase levels of D-dimer in case group, mean 0.31±0.403 compared with control group, mean0.133±0.043 (P < 0.003), while no significant differences in fibrinogen levels between patient’s group and control group (P > 0.05).
The PSGL-1 allelic frequency of A, B and C alleles was found to be 85%, 14% and 1% respectively in control group, while in patients there was difference in B allele frequency compared with control samples which is statistically significant 26% versus 17.4% (p < 0.01). The AB genotype was significantly more frequent among case group than in control group40.6% versus 20% (p < 0.02)
C > T substitution sequence chromatogram in case group, shown by GATC Viewer program. Bio Edit multiple sequence alignment showed first and second the gene sequence from gene bank, but no mutation at the protein level in the same position detected by Using chimera software. |
