Abstract: The aim of this study was to detect the presence of Helicobacter pylori (H. pylori) in fresh milk samples in Khartoum State in the period between March and November 2018. Total of 50 fresh milk samples were taken from cows (18 sample) , goats (18 sample) and sheep (14 sample). 39 sample were collected in the morning, while 11 were collected in the evening using sterile containers. All milk samples were cultured on Columbia blood agar media for isolation. DNA extraction by guanidine chloride method and multiplex PCR were applied to these samples to detect H.pylori glmM and 16s RNA genes. The results showed that out of 50 samples, no growth was seen on culture media, and there is no gene detected by PCR technique (0%). It is suggested that H. pylori is not present in animal sources and transmission of H. pylori from animal sources such as milk is not a primary factor in infection related to this bacterium in humans. More research work is needed to validate these results.
